一般螢光的訊號很弱, 所以我建議用二抗接biotin
中間再用avidin-biotin將訊號放大, 最後先用DAB這種可見光的substrate來染,
若有染上馬上就能看見
另外, fix後的組織都會用citrate buffer, 100度煮個10~20分鐘, 讓抗原恢復
