(abstract)
主要是在證明phoP gene所調控的一些基因與Macrophage的死亡有關,
利用wild type與phoP::Tn10 mutant去感染細胞株
再利用microarray去分析這兩種細胞之間所表現的基因有哪些相同與不同處
(introduction)
S. typhimurium can kill macrophage by a caspase-1-dependent mechanism
提到了許多受phoP 影響的基因,包括:
1.resistance to anti-microbial peptides by chemically modifying LPS
(pmrA/B,pmrE,pmrF,pagP)
2.magnesium transport to play resistance to bile (mgtA,mgtCB)
3.entry into epithelial cells (SPI1)
--SPI這個在這篇PAPER常提到(包括:SPI2 type III secretion machinery...等等)
那我們的細胞到底對這些受phoP調控的基因會產生什麼樣的反應呢?
因此我們才長是利用microarray去偵測到底細胞受野生株和突變株感染之後,
此兩者之間細胞的基因表現主要的差距是在哪裡呢?
(materials and methods)
1.bacterial strains and growth conditions
2.tissue culture infections and bacterial replication assays
3.RNA isolation, labeling, and hybridization(這邊也不懂)
4.data analysis(我們最不懂的地方)
5.Northern(對照MICROARRAY的結果罷了,有印證的效果吧,只不過是從RNA的角度下手)
6.cell death assays
-LDH release assay -TUNEL-fluorescence-activated cell sorter assay(這個不懂)
(results)
1.phoP::Tn10 mutant Salmodella replicate in U-937s
--證明仍可以在細胞內複製!
2.expression profiles of Salmonella-infected Vs. uninfected tissue culture cell
--針對有感染與沒有感染隻細胞在基因表現程度上有何不同!
3.a phoP::Tn10 mutant strain elicits expression of many of the same macrophage
mRNAs as wild-type Salmonella
--證明突變株感染與野生株感染的巨噬細胞,在許多phoP可能影響之基因的表現上仍舊一樣
4.a phoP:Tn10 mutant Salmonella fails to elicit host cell induction of some
cell death and cell cycle genes
--雖然突變株與野生株感染的細胞在許多基因的表現上仍舊一樣,但是我們也發現到
在phoP::Tn10 mutant-infected macrophage上,有許多和細胞死亡與細胞週期有關的基
因表現量比受野生株感染的表現還少,舉了許多基因當作例子,關於這邊我們可能要思考
一下該如何表達才好!
5.Northern analysis of mRNA samples
6.the phoP::Tn10 mutant Salmpnella kills fewer tissue culture macrophages than
wild-type Salmonella
--我們在microarray發現到,突變株感染的細胞在與細胞死亡以及細胞週期有關的基因表現
量上較低,因此我們藉由cell death assay也證明受突變株感染的macrophage死亡的數目
較少,因此兩者之間有其正相關的存在
7.phoP is important for killing human PBMCs
--證明從人的周邊血液取出的macrophage,與之前實驗中的細胞株,對此結果有一致的傾向!
