這題的文法其實是在考「Whether (or not)… 」(不論…/是否…) 。
Whether John is good or bad, he is my friend.
上面這句對大部分的人都沒有問題，就是
「不論約翰是好是壞，他都是我朋友」。
若在比較文言或文學的文章裡，如果是一般現在式，則動詞可改成「原形動詞」。
但這種用法在口語或是現代散文非常少見。
Whether John is good or bad, he is my friend.
= Whether John be good or bad, he is my friend.
然後，這種表示讓步的whether/if/no matter是可以省略和倒裝的，如下：
Whether John is good or bad, he is my friend.
= Whether John be good or bad, he is my friend.
= Be John good or bad, he is my friend.
再舉一例：
Whether the test is easy or difficult, I will pass it.
= Be the test easy or difficult, I will pass it.
之前在高中參考書有看過這麼一句經典名言：
Be it ever so humble, there is no place like home.
這句其實就是跟上句一樣，也是減化倒裝的結果↓
Be it ever so humble, there is no place like home.
= No matter how humble it is, there is no place like home.
回到你的問題來，原句應該是↓
Whether it is work or play, we have to try our level best.
上面的whether被省略掉，然後is被改成原形動詞「be」，倒裝移到句首，
就成了題目的這句：
Be it work or play, we have to try our level best.
