在以前，還是可以做作的啦，只是運氣成份“超大“，首先
1.protein來源，做 N-ter sequencing，一般而言，有辦法定出6個amino acid就夠了。
2.有Wobble hypothesis的觀念是正確的，所以人們也很天才的把所有的可能的forward
primer都作出來，作什麼呢……RT-PCR，那reverse的primer要用什麼呢……oligo dT
N-ter--------------------------C-ter

5'------/------------------------------/-----/--------3'
^^^^^^ ^^^^^ ^^^^^^^^
5'-UTR 3'-UTR poly-A tail
↓ ↓
理論cDNA:5'------------------------------/-----/-----
所以理論cDNA會含3'-UTR與一部份的poly-A tail
就到此為止而言，需要相當的運氣……
而後就是cloning的東西……，可以作到sequencing為止。

我想他指的是相同gene的產物，但是是不同的protein，例如：經過alternative splicing
的mature mRNA，是來自不同exon接起來的……(我自已想，應該是在RNA level)

這個我認為是找siRNA才會用到的方法，由於miRNA往往是自然存在於生物體中，從
genomic DNA transcript而來，所以我個人較偏向是設計siRNA時所採用的方法。
