演講摘要: Pharmaceutical development requires chemical engineering principles
and approaches from laboratory process development to scaling up. The
presentation focuses on two major segments in developing active
pharmaceutical ingredients (API drug substance): reaction and
crystallization. Conventional batch processing as commonly used in
pharmaceutical synthesis often encounters challenges when scaling up. As an
alternate technology in pharmaceutical process development, continuous
processing may pose a solution addressing issues with safety, quality or
economics which may be magnified upon scale-up. In particular, three
processes will be highlighted: (1) controlling reaction purity profiles, (2)
managing a reaction exotherm and reagent stability, and (3) converting a
cryogenic synthesis to an energy-efficient noncryogenic operation.
Crystallization of pharmaceutical intermediates or APIs governs the quality
of the compound developed; specifically for APIs the desired polymorphs and
powder properties have great impacts on their suitable use in drug substance
application. Examples of crystallization and particle engineering will be
discussed involving processing techniques to tailor crystal phase, crystal
morphology, particle size, as well as filterability and powder property in
isolation.
