[求救] 新手求步驟
Construction of the pPre-dsRBD plasmid:
Vector: pBase digested with Pst I and Bgl II, and gel-purify 2.7 kb
Insert: pCR-dsRBD digested with Pst I/Bgl II, and gel-purify 227 bp
dsRBD of pET47b-dsRBD plasmid (kindly provided by Scott Showalter, PLoS One
7:e51829, 2012) was PCR-amplified by dsRBD-U1 and dsRBD-D1 primers, then
cloned into pCR T7/CT TOPO named pCR-dsRBD
dsRBD-U1 (39mer): 5'-AACTGCAGACCGCGTAGTCCGGTTCGTGAACTGCTGGAA-3' (5'-terminus
has a Pst I site)
dsRBD-D1 (39mer): 5'-TTAGATCTTGCTATTCGGAACCTGCGGCTGATTTGCTTT-3' (5'-terminus
has a Bgl II site)
最近要做這個
已經先做好了一罐
pCR-dsRBD 0.062ul
和
pBase 0.128 ul
但是毫無頭緒再來要做什麼
救命~
