[求救] RNA的OD值過高
最近在試用一個新的kit
主要是從全血同時將DNA和RNA粹取出來
步驟和抽DNA的時候類似
如果要DNA或RNA其中一種
就在利用DNase或RNase去除不要的東西
結果出來後 利用nano-drop測其濃度
DNA或RNA的260/280的值都奇高無比
DNA大約是4 RNA甚至有高到9
我大概查一下
似乎是因為酒精殘留
但是我已經在hood風乾2個小時了
值還是這麼高
我們實驗的目標是RNA
因此我主要希望是RNA夠純 才可以做接下來轉cDNA的步驟
我看到網路上的資料有提到RNA的260/230的值也要留意一下
建議260/230的值要高過1.8 而我的260/230都不到0.1...
搜尋一下是因為有鹽類等的干擾
但是不曉得要怎麼debug.... Orz
這次我試的sample是用paxagen kit fix過RNA的全血
但是他已經放了快5年了.... @@
我不曉得這樣的sample是不是也會影響結果?
麻煩板上的高手幫我debug >_<

以下是我的實驗步驟:
AquaPreserve/Prosink皆為這個kit的reagent
1. Pre-warm the AquaPreserve solution to 22-37℃
2. Add 250 μl AquaPreserve to 250 μl whole blood in a 1.5 ml microfuge tube
3. Vortex until frozen blood-AquaPreserve thaw completely
4. Add 125 μl Prosink and vortex 1-2 min
5. Incubate 22℃ for more than 30 min
6. Centrifuge 14,000 xg for 10 min to pellet the proteins
7. Transfer 0.5 ml supernatant to a new 1.5 ml tube
8. Add 0.4 ml isopropanol to (7) and shake/vortex for 30-60 sec
9. Centrifuge at 14,000xg for 10 min to pellet the DNA/RNA
10.Decant to discard supernatant and gently add 50% isopropanol or 70%
ethanol to fill up the tube
11. Decant to discard the alcohol solution
12. Tap the tube on a paper towel to remove residual liquid
13. Leave it upside down to air dry the DNA/RNA pellet for 5-10 min
14. Add 100 μl deionized water to the DNA/RNA pellet and vortex to
solubilize the DNA and RNA
15. Add DNase and incubate 22~37℃ for 20 min
16. Incubate at 65℃ for 30 min to inactivate DNase
