[方法] ATP solution
各位板友好
最近需要配製ATP solution for cell culture stimulation
查過sigma catalog有找到ATP anhydride powder & cell culture tested
solubility in water = 50mg/mL 換算成molar concentration約100mM
google找過方法 也想過要不要買現成的ATP solution(大多是分生用途)
因為本身實驗用量較大(每組實驗至少需50 nMole 如果買分生用的1 ml/vial用太凶)
有查到資料說ATP在非生理pH值(6.8~7.4)不穩定 所以solution要調pH
而且是細胞用 理應配在PBS裡? 過0.22 um filter會不會被吸光光?
不知道有沒有板友能夠建議較好的方式可以prepare solution? 彼此討教一下:)
ps.是要測試inflammasome相關的實驗 paper的ATP working conc. 5mM
我目前初步的想法是 配在LPS-free distilled water
因為一般配PBS的二次水有太多endotoxin 可能造成不必要之background
1g ATP powder in 20ml water = 100mM stock
假設過0.22um-filter沒有問題的話 調好pH再過濾 分成vial凍-20度
實驗時以20x dilution 每1ml culture就要加50ul aqueous solution 會不會太多?
怎麼想好像都怪怪的 目前手邊也沒有ref可以參考 不知道大家有沒有類似經驗呢?
