[求救] Bradford protein assay問題

我的protein lysis sample裡有1% SDS 所以sample用純水稀釋了100倍以後就加上Comassie solution做bradford assay 然後發現顏色是鮮藍色的再做了blank blank就是0.01% SDS 再加純水還有Comassie solution 而standard就用20ug/ml的BSA, 溶在純水中第一個standard用0.02% SDS 稀釋之後的都用0.01% SDS稀釋 (serial dilution的方法) 所以standard是 10, 5, 2.5, 1.25, 0.625 跟 0.3125 ug/ml 度595nm時 blank的讀數比standard還有強 standard的數都變負了@@" 而sample都差不多是零我在assay中是不是有什麼做錯了? 在manual裡是說可以測sample裡有0.1%或以下的SDS啊@@" 或者是有什麼其他的方法去測protein濃度的方法嗎? 謝謝!!!! -- ※ 發信站: 批踢踢實業坊(ptt.cc) ◆ From: 144.214.50.151
