[求救] CytoTrap 的 agar plate
最近做 yeast CytoTrap 的實驗，依照廠商的 Protocol 配 Agar Plate
Plate 偏軟，用 beads塗或 loop 劃三區 agar 都會破掉，
本來以為是配錯了，重新配二、三次（確定沒秤錯或少加），還是偏軟。
下面是廠商的配方，想請問有配過的人，Agar是加多少？
Synthetic Glucose Minimal Medium [SD/Glucose (–UL)] (per Liter)
1.7 g of yeast nitrogen base without amino acids
5 g of ammonium sulfate
20 g of dextrose (Glucose)
add 17 g of Bacto agar for SD dropout agar plates (use DIFCO agar)
Adjust the total volume to 900 ml with ddH2O
Autoclave for 15 minutes at 121°C, cool to 55°C
Add 100 ml of the appropriate filter-sterilized 10× dropout solution
這裡是加 17 g 的 Agar，但是卻軟軟的
一般我們在配 LB plate 時也頂多加 15 g 的 Agar，可是就夠硬了
到底是為什麼呢？
