[方法] 請問有沒有人做過peptide binding然後作Flow?
是這樣的 目前是研究依一個secreted protein
覺得它應該會bind 某receptor
老師希望可以找具有此receptor的 cell population
所以 目前想 把這 protein去和cell binding
然後染此protein以及幾個surface marker
上flow看一下這個ligand會bind哪種cell
請問有人做過類似實驗嗎
目前是 a. protein binding RT 1hr
b. 2.4G2 blocking on ice 15min
c. 一抗(染此protein) RT 1hr
d. 二抗 RT 30min then surfacr marker Abs 30min on ice
實驗結果 減加protein binding沒差異 都沒有binding
有學長認為不要室溫 receptor會endocytosis 所以bind不到
但 記得好像on ice receptor和ligand應該不會bind吧 ??
有先進有好建議嗎
目前是想縮短染色時間 感覺染太久也對細胞不好 死亡率會提高
